Act 2 is when the difficulty really skyrocketed for me. I beat the first boss on my 6th or 7th overall run. A few more attempts, then I beat 2 + 3 on the same go. (3 may be my favorite boss fight of all time so far). And then I hit a huge wall and died like 20+ times before beating boss 4. And from there it took another big chunk until I finally got to the last biome.
But goddamn. This game is constantly, endlessly surprising and delighting me. It's truly incredible.
Boss fight 3 was mindblowing, haha. Biome 4 was a wall for me as well, but I'm mid-run through Biome 5 and it's going smoothly now.
One tip I just learned that frustrated me, btw. Spoiler tagging in case you want a pristine playthrough, but it has to do with collecting all the loot in a biome:

You need to get the Blade Enhancer artifact in order to break open the yellow barriers that pop up here and there. I never bothered to buy it because melee is so strong anyway.
I learned that today and it got me some key loot. I beat the game last night (so fucking good) and was trying to do a perfect run, thorough through the whole game. I was smacking around Boss 2 after building up 250% integrity and endless good artifacts and parasites and it fucking crashed on me
